Ruling Ectoplasm and Power-spray

Basically, If my opponent's Dusknoir LV X uses Ectoplasm, can I Power-Spray it to cancel it?

No, you can only play power-spray on teh opponets turn, not your own.

It's so easy to just read a card first, you know. As Foxtrot states, Power-Spray (Just like Alakazam MT) can only be used on your OPPONENT's turn.

but if is the dusknoir SF#1 uses its power and kill himself on YOUR OPONENTS TURN?

Ectoplasm only works when it gets KOed from attacks so theres no need to worry here.
